Evaluation of Novel Compound Benzoylphenyl-Indole-Carboxamide Effect on Hyperlipidemic and Hyperglycemic Rat

Abstract : Hyperlipidemia is one of the most risk factors involved in the development of cardiovascular disease. As a consequence of hyperlipidemia treatment, a high demand for new oral antihyperlipidemic drugs is required. The present study was designed to investigate the potential effect of a novel compound N-[4-benzoylphenyl]-1H-indole- 2-carboxamide on diet-induced hyperlipidemic (prepared by mixing cholesterol 0.5%, 0.25 % cholic acid, 20% fat and 2% corn oil with standard powdered animal food for two months) , triton-induced hyperlipidemic (by intraperitoneal injection of Triton WR-1339 250 mg/kg body weight) and alloxan-induced hyperglycemic rats (150 mg/kg body weight injected intraperitoneally). Lipid profiles [Total cholesterol (TC), Triglyceride (TG), Low Density Lipoprotein (LDL) and High Density Lipoprotein (HDL)] in all groups showed significant (p < 0.05) changes (±) before treatment. After 24 hrs of treatment with N-[4-benzoylphenyl]-1H-indole-2-carboxamide (15 mg/kg body weight), significant decrease (p <0.05) in serum cholesterol level 60%, , triglyceride 33-45%, LDL 32% and increase HDL 58% levels. Nevertheless, antihyperlipidemic fenofibrate - used as standard reference - showed parallel results to the novel compound. In addition, body weight of diet-induced hyperlipidemic rats decreased about 20%. On the other hand, the novel compound showed no significant effect on glucose level of hyperglycemic rats. In conclusion, the results indicate that N-[4-benzoylphenyl]-1H-indole-2-carboxamide possesses significant antihyperlipidemic activity and may conserve a promising potential effect in the treatment of hyperlipidemia correlated to heart diseases.

Effects of Hawthorn on HbA1C and lipids levels in Jordanian diabetic patients (Type2)br/>

Abstract : The current research was intended to elucidate the effects of supplementation of hawthorn on the plasma level of glucose, glycosylated hemoglobin HbA1C and lipid emic parameters which include triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL) and high density lipoprotein (HDL) among type 2 diabetic patients. Fifty five diabetic type 2 patients were encountered and received 3 grams dose of hawthorn powder administered orally in the form of 500 mg capsules 3 times daily at breakfast, lunch and dinner meals for 12 weeks. Measurements of fasting blood glucose, HbA1C and lipid parameters before and after treatment showed significant effects at P < 0.05 were recorded due to hawthorn treatment. In brief, glucose level dropped from 223.6 mg/dl to 186.34 mg/dl HbA1C decreased from 8.5% to 7.2%, TG was reduced from 235.5 mg/dl to 160 mg/dl, TC was declined from 310 mg/dl to 187.6 mg/dl, LDL was decreased from 155.2 mg/dl to 115.5 mg/dl and lastly high HDL increased from 52.4 mg/dl to 69.2. mg/dl. It was concluded that hawthorn may be one of the future optimizing medicinal plants that shows dual effect against hyperglycemia and hyperlipidemia.

Effect of Cinnamon Administration on Fertility of Normal and Diabetic Male Rats

Abstract : Background: Male infertility is one of the most problem in the word Diabetes is one of the chronic metabolic syndrome that was found to have direct effect of testicular malfunction. Our study aimed to evaluate the enhancement effect of cinnamon bark powder on fertility parameters in normal and diabetic male wistar rats.
Material and Methods: 40 male wistar rats were divided into two sets: normal and diabetic rats. Normal rats received either distilled water or 80 mg/kg/day of cinnamon, and diabetic rats received either distilled water or 80 mg/kg/day of cinnamon. Cinnamon powder was suspended in distilled water. After four weeks all groups were euthanized and analyzed for testosterone, epididymis weight, sperm count, motility, and morphology with proper technique.
Results: The presented data describe that cinnamon as powder exerted significant changes on fertility parameters.in both normal and diabetic rats including testosterone, epididymis, total sperm count, motility, and sperm morphology (normal sperm count, abnormal head sperms, abnormal tail sperms). Also it was noticed that diabetic rats gain higher beneficial improvement changes in fertility parameters than normal rats.
Conclusion: Ingestion of cinnamon bark found to improve significantly infertility parameters in both normal and diabetic male rats which may give promising solution to the problem, however more effective on diabetic than normal rats.

Effect of rosemary on fasting blood glucose, hemoglobin A1c and Vitamin B12 in healthy person and Type 2 diabetic patients taking glucomid or/and metformin

Abstract : Background: Diabetes mellitus (DM) is risky threats to human health nowadays, and various trials of treatments were announced including chemical and drugs extracted from medicinal plants. Rosemary plants were shown to have antidiabetic and antilipidemic effects. Aims and Objective: Evaluate and investigate the effect of Rosemary administration on hemoglobin A1c (HbA1C) and Vitamin B12 in healthy and diabetic patient who intake glucomid or metformin. Materials and Methods: Forty individuals with Type 2 diabetes of both genders and 10 healthy persons of age 40 years or older were recruited for participating in this study. Diabetic patients were divided into two groups: The first one was taking only glucomid, and the other group was taking both glucomid and metformin together. Rosemary was supplemented as 500 mg powder-encapsulated. All participants were asked to intake 3 g/day (2 capsules at breakfast, lunch, and dinner)and be conducted for 8 weeks. Results: Our data and results showed that rosemary has potentially significant effects on fasting blood glucose (FBG), HbA1c and Vitamin B12 in both healthy and patient’s participants. In healthy persons, level of Vitamin B12 was improved 29%, with some effect on HbA1c and FBG (5%, 14%, respectively). In diabetic patients, who were uptaking glucomid only, a significant reduction about 16-18% in both FBG and HbA1c, with a prominent increase in Vitamin B12 level (25%). On the other hand, patients who were receiving both glucomid and metformin responded more effectively to rosemary in FBG and HbA1c (23%) with less increase in Vitamin B12 (10%). In comparison between the levels of Vitamin B12 before rosemary consumption, data indicated that Vitamin B12 level in diabetic patients was 40% less than healthy persons notifying that diabetic patient may suffer from Vitamin B12 deficiency. Conclusion: The present results declare that rosemary can exert significant effects on improving levels of FBG, HbA1c, and Vitamin B12 in both healthy persons and diabetic patients involved in antidiabetic medication.

Effect of Fluconazole and Isobutyl Methylxanthine (IBMX) on Temporal Expression of CgCDR1 and CgCDR2 Genes in Candida glabrata

Abstract : Hospital. The relative expression of resistant CgCDR1 and CgCDR2 genes in this clinical
isolate in response to fluconazole in the presence and absence of phosphodiesterase
inhibitor IBMX (3-isobutyl-1-methylxanthine) was characterized. Dose analysis showed
that CgCDR1 was expressed in response to 8, 16, and 24 μg/ml of fluconazole but not to 32
and 40 μg/ml, whereas CgCDR2 was expressed in response to all fluconazole
concentrations even in untreated cells but to a lesser extent. Temporal analysis of relative
mRNA expression was investigated against the different concentrations but here,
results of 24 μg/ml fluconazole will be only represented. CgCDR1 expression appeared
after 2, 4, or 6 hrs in response to 24 μg/ml fluconazole, whereas CgCDR2 expression was
maintained equally throughout all time intervals and was observed in untreated cells as well.
Stabilization of endogenous levels of cAMP by addition of 3mM IBMX further increased
expression levels of CgCDR1 and CgCDR2 (to a lesser extent) in time points that have
exhibited an increase in expression in response to fluconazole. These results suggest
differential upregulation of CgCDR1 and CgCDR2 in fluconazole resistant of C. glabrata
and may implicate a possible role of cAMP in resistance mechanism to fluconazole in C.
glabrata.

Effect of Green Coffee and Orlistat on Obese Individuals

Abstract : The present result data represents evaluation of some vital physiological parameters in obese male human after uptaking green coffee beans or/and anti-obese orlistat drug. Parameters of blood glucose, lipid profile, blood pressure and some physiological minerals in addition to vitamin B12 are implemented. Green coffee beans alone showed significant effect on most studied parameters. The results show reduction about 15-21% in fast blood glucose and lipid parameters (cholesterol, triglyceride, and LDH) in addition to 32% increase in HDL. On the other hand, Orlistat showed less effect on these parameters about 8-13%. No noticeable effect of either green coffee beans or orlistat on blood pressure. Moreover, among physiological minerals, Mg increased significantly (30%) due to green coffee uptake compared to similar effect of orlistat, whereas other minerals (Zn, Ca, Mg) and vitamin B12 were not affected in the presence of either green coffee or orlistat. Combination of green coffee and orlistat didn’t exert any additive effect on all parameters. In conclusion, green coffee may be recommended to modify some physiological parameters in human.
